检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠 TRANCE 抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的 TRANCE 与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物 TMB,避光显色。颜色反应的深浅与样本中 TRANCE 的浓度成正比。加入终止液终止反应,在 450 nm 波长(参考波长 570 - 630 nm)测定吸光度值。

TNFSF11 分子靶点综述
肿瘤坏死因子相关激活诱导因子(TRANCE),又名肿瘤坏死因子配体超家族成员11(TNFSF11)、核因子κB受体活化因子配基(RANKL),是TNF超家族成员。成骨细胞、CD4+和CD8+T细胞、巨核细胞、支气管和肠道上皮细胞等都可表达TRANCE。TRANCE在免疫应答和骨形态发生中起核心作用。在骨形成和重塑过程中,成骨细胞表达的TRANCE与M-CSF一起诱导破骨细胞形成和活化,导致骨吸收,进而导致随后的骨沉积和重塑。TRANCE同样在病理性骨质疏松中起作用。活化的淋巴细胞和成骨细胞不恰当表达TRANCE就会导致关节和骨头的过度吸收与沉积。过量的TRANCE与许多退行性骨疾病,如类风湿性关节炎、银屑病性关节炎有关。
